    Sterilization test against human flu virus

    Material


    Virus: Influenza virus
    Virus titer: 4.0 x 10^8pfu/ml (4.0 x 10 to the 8 pfu/ml)
    Cell :CaCO2 cell for Infuluenza virus
    Method for counting virus: Plaque method by CaCO2

    Test method

    1. Kilvi original(400ppm) was dilute by pure water. Dillution from 10ppm to 100ppm
    2. To put 50ul of virus into 450ul of above diluted water . And we see the result in 1-5minutes.
    3. After that, putting 10% Na2S2O3(Sodium thiosulfate) to stop the react.
    4. Kilvi water including virus is dilluted in 10 times by PBS. And then 0.1ml each is put into 4 well CaCO2. Temp 37 centigrade, 60minutes
    5. After that CaCO2 is put into 0.8% medium and wait 72 hrs After that it fixed by Methanol. And then we put 20% ethanol with 0.1% cystal violet to see Virus titer.
    6. This experiment is done by twice.

    Result

    If flu virus contact with Kilvi more than 3 min, most of them are sterilized.
